Formulation and Evaluation of Double-layered (Matrix and Drug-in-adhesive) Transdermal Patches of Diclofenac Diethylamine: In vitro and ex vivo Permeation Studies

Abstract: Background: The objective of this current study was to fabricate and characterize the sustained release double layered transdermal patches Diclofenac Diethylamine using hydrophobic acrylic polymer Eudragit RL 100 hydrophilic Polyvinyl pyrrolidone K-30 in combination as first layer matrix type patch pressure sensitive adhesive Duro Tak 387- 2510 second drug-in-adhesive provide anti-inflammatory effect. Materials Methods: Solvent casting method employed prepare over backing membrane. technique were attempted create desirable features adhesive. drug-polymers compatibility studies examined by Infrared spectroscopy. A full-thickness excised abdominal rat skin sample used perform permeation Franz’s Diffusion Cell. Results: Drug-polymers revealed no evidence interaction formulations showed satisfactory physicochemical-mechanical characteristics. It found that profiles affected proportion enhancer used. optimized formulation MF3 maximum vitro ex vivo percent cumulative drug permeated i.e. 87.28 ± 3.88% 81.95 2.64% respectively, with flux 12.96 0.51 μg/ cm2/h permeability coefficient 1.09 0.45 cm/h, 24 hr. Also, suggested dimethyl sulphoxide exhibit more promising results enhancer. Conclusion: concluded an ideal would serve best carrier effect enhancement Keywords: Transdermal patches, Double technique, Hydrophobic polymer, Hydrophilic Permeation enhancer, Anti-inflammatory.
